Background Information: | PathHunter® U2OS CD33 Internalization Cell Line is engineered to co-express the ProLink™ (PK) tagged CD33 receptor, and an Enzyme Acceptor (EA) tag localized to the endosomes. Ligand (e.g. Sialic Acid, anti-CD33 antibody) binding to the receptor stimulates receptor internalization, resulting in colocalization of the receptor-PK with the EA in the endosomes, and forcing complementation of the two β galactosidase (β-gal) enzyme fragments (EA and PK). The resulting functional β-gal enzyme hydrolyzes substrate to generate a chemiluminescent signal. This stable modified cell line is provided as two vials of cryopreserved cells. |
