PathHunter® PD-1 (SHP1) Signaling Bioassay Kit

The PathHunter PD-1 (SHP1) Signaling Bioassay Kit provides an easy-to-use cell based assay to measure drug potency and detect neutralizing antibodies. This bioassay asseses ligand (e.g. Anti-CD47 Antibody (Ab-2; Clone B6H12.2)) based on activation of PD-1, SHP1 Checkpoint activity via detection of SH2 Recruitment. The included cell line overexpresses ProLink-tagged PD-1, SHP1 and EA-tagged . Activation of PD-1, SHP1 stimulates the recruitment of and produces EFC signal. Bioassay kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, control agonist, detection reagent, and assay plates.

The graph is an example of a typical dose-response curve for the PD-1 Signaling Bioassay, generated using the protocol outlined in this user manual. The data shows a potent, dose-dependent inhibition of PD-1 activation in Jurkat cells with an anti-PD-1 antibody.
Format
See User Manual for Product Configuration Details. The purpose of the control ligand included in the Bioassay Kit is for verification of assay performance relative to the data shown in the Certificate of Analysis for that kit. This control ligand is not intended for use as a reference standard for validation studies and lot release applications. Typically, a reference medicinal product (RMP) that has been pre-defined by the customer as an internal reference standard is used for those applications. The RMP is not provided with the bioassay kit or supplied by Eurofins DiscoverX.
