PathHunter® U2OS PDGFRb Bioassay Kit

The PathHunter U2OS PDGFRb Bioassay Kit provides an easy-to-use cell based assay to measure drug potency and detect neutralizing antibodies. This bioassay asseses ligand (e.g. Recombinant Human PDGF-BB) based on activation of PDGFRb Kinase activity via detection of SH2 Recruitment. The included cell line overexpresses ProLink-tagged PDGFRb and EA-tagged . Activation of PDGFRb stimulates the recruitment of and produces EFC signal. Bioassay kits are a convenient, ready-to-use format which contains all materials needed to run the assay, including single-use vials of cryopreserved cells, cell plating media, control agonist, detection reagent, and assay plates.

Cells expressing human ErbB1 receptor were seeded in Cell Plating 17 Reagent and incubated with increasing concentrations of an anti-ErbB1 MAb, keeping agonist, human EGF, constant at the EC80 according to the protocol. ErbB1 Bioassay Kit is able to detect a dose dependent neutralization of receptor signaling as increasing concentrations of anti-ErbB1 mAb bind directly to the receptor and block EGF ligand signaling.
Format
See User Manual for Product Configuration Details. The purpose of the control ligand included in the Bioassay Kit is for verification of assay performance relative to the data shown in the Certificate of Analysis for that kit. This control ligand is not intended for use as a reference standard for validation studies and lot release applications. Typically, a reference medicinal product (RMP) that has been pre-defined by the customer as an internal reference standard is used for those applications. The RMP is not provided with the bioassay kit or supplied by Eurofins DiscoverX.

Short term (<24 h): Store at -80°C; Long term (>24 h): Store in vapor phase of liquid nitrogen.
