Camptothecin

Camptothecin is a cytotoxic quinoline plant alkaloid that inhibits DNA and RNA synthesis by specifically inhibiting the DNA topoisomerase I enzyme. Shows great anti-tumor potential in the clinic by inducing DNA breaks and apoptosis, but shows in-vivo cytotoxicity. Derivitives of camptothecin have been developed and are used in chemotherapy, such as toptecan and irinotecan.

Cells were plated in a 384-well plate and incubated overnight at 37°C/5% CO2 to allow the cells to attach and grow. Cells were then stimulated with a control compound, using the assay conditions described below. Following stimulation, signal was detected using the PathHunter Detection Kit according to the recommended protocol.
