LCK Inhibitor

LCK (lymphocyte-specific protein tyrosine kinase) inhibitor is a pyrrolopyrimidine that block the activity of two forms of Lck kinase, Lck (64-509) and Lckcd, and has been shown to inhibit T cell receptor-stimulated IL-2 production. The LCK protein is a member of the Src-family of non-receptor protein tyrosine kinases and plays a critical role in the initial steps of T cell receptor signaling that trigger the production of cytokines.

Cells were plated in a 384-well plate and incubated overnight at 37°C/5% CO2 to allow the cells to attach and grow. Cells were then pre-incubated with inhibitor, followed by stimulation with the control agonist using the assay conditions described below. Following stimulation, signal was detected using the PathHunter Detection Kit according to the recommended protocol.
