Thapsigargin

Thapsigargin is a naturally occurring, cell-permeable plant sesquiterpene lactone that non-competitively inhibits the sarco/endoplasmic reticulum calcium ATPase (SERCA, IC50 ~2-80nM). Effectively increases intracellular calcium levels upon SERCA inhibition, leading to cell activation, increased histamine release from mast cells, and increased cancer cell proliferation. Acts as an anti-inflammatory and induces autophagy in-vivo.

Cells were plated in a 384-well plate and incubated overnight at 37°C/5% CO2 to allow the cells to attach and grow. Cells were then stimulated with a control compound, using the assay conditions described below. Following stimulation, signal was detected using the PathHunter Detection Kit according to the recommended protocol.
