Background Information: | Wortmannin is a natural product that potently inhibits PI3K with anti-inflammatory and immunosuppressant effects in vivo. It has been used extensively in the study of insulin-regulated events in a variety of cells. It was also used to study the activation of PI3K by other growth factors, such as platelet-derived growth factor. Wortmannin was reported to block insulin-stimulated expression of lipogenic genes, such as Srebp-1c and Acc. It also blocked insulin-stimulated glycogen synthase, S6K, Akt, and GSK-3 activities. Interestingly, it has been reported that Wortmannin also inhibited the activity of smooth muscle myosin light-chain kinase. Wortmannin is a naturally occurring steroid metabolite that potently inhibits phosphatidylinositol 3-kinases. Wortmannin also inhibits PLK1, PLK3, MTOR, DNAPK, PI4Ks, MLCK, and MAPK at higher concentrations.
