For therapeutic antibodies, binding to Fcγ receptors and complement component 1q (C1q) are related to immune responses such as Antibody-dependent cell cytotoxicity (ADCC), Antibody-dependent cell phagocytosis (ADCP), and Complement dependent cytotoxicity (CDC). Also, binding to FcRn receptor is reported to affect the antibody half-life in serum. Therefore, characterization of antibody binding to Fc receptors or to complement is highly recommended by health authorities including FDA, EMA, ICH, and NMPA. The use of Surface Plasmon Resonance (SPR) to analyze molecular binding is widespread due to its high sensitivity, real-time monitoring, and label-free detection and can measure not only the steady-state binding affinity but also the kinetics constants.
